DT4DDS - Characterization of Errors and Biases in DNA Data Storage

In this study, the errors and biases present in state-of-the-art data storage workflows were characterized and a digital twin was constructed to enable rational design of data storage experiments. Experiments encompassed synthesis by electrochemical synthesis and synthesis by material deposition, PCR, aging, and sequencing-by-synthesis via Illumina iSeq 100. Both the error patterns and coverage biases were comprehensively quantified by sequencing experiment samples along the state-of-the-art workflow.
