Here, we use elongation velocity analysis showed that acute loss H3K4me3 caused a general transcriptome-wide attenuation of elongation velocity in cells. Overall design: In order to examine and monitor the effects of H3K4me3 on newly synthesized RNA rates genome-wide from actively transcribing RNAPII, we performed the modified transient transcriptome sequencing (TTchem-seq) and DRB/TTchem-seq with spike-in strategy on degron cells.
