Concurrent alterations in cell cycle regulator genes have shown to contribute to resistance to targeted therapy in NSCLC. Here we analyzed the functional consequences of CDK4 and CDK6 protein overexpression in EGFR mutant NSCLC NCIH1975 cell line-derived xenografts, identifying key transcriptional signatures that contribute to E2F-dependent G1/S cell cycle progression and proliferation, DNA repair, G2/M checkpoint and reduction of NfkB, EMT and apoptosis pathways.
