E. coli ET5, expressing Gleobacter rhodopsin from pKJ606-GR plasmid, exhibits a rhodopsin-based phototrophic metabolism. ET5 was isolated after 88 days of chemostat culture with constant illumination with limited glucose as the sole carbon source. This study is to elucidate how cells optimize their newly acquired phototrophic metabolims by the rise of mutations.
