资源简介:
The goal of this study was to quantify transcriptomic differences between sphereoids made from bronchial epithelial cells at different stages of their lifecycle. These differences in their transcriptome were then compared with the transcriptomes of their progenitor cells, normal human bronchial epithelial cells (NHBEs). The samples of this study fall into several major groups. The first are the normal human bronchial epithelial progenitor cells (NHBEs); these are the cells used to make the Anthrobots, airway organoids. There are several passages collected, denoted by "P + No." and these are the times the cells were passaged following thaw from the original frozen stock. The next set collected was the RNA from Anthrobots at various stages of their lifecycle. The RNA of the day 0 bots was collected immediately upon their being dissolved from their Matrigel suspension. The following day 10, day 25 bots were collected 10 and 25 days post-Matrigel respectively. The Bridge Bots were where we dissolved an entire well into a 60mm dish to allow for better aggregation between bots; the bots are capable of aggregating to form larger organoids, and the normal method is to divide the contents of a single well among two 60mm dishes to discourage aggregation
