CMV-specific CD8+ T cells from HLA-A*0201 kidney transplant recipients were stimulated and isolated using A*0201 dextramers loaded with restricted pp65 epitope (costimulation performed with aCD28/aCD49 at 1 ug/mL each; total stimulation time 6 hours, no BFA/monensin used). Cells were sorted into individuals wells of a 96-well plate (one cell per well) and a single-cell transcriptional library generated using the BD Precise Assay per manufacturer protocol. Sequencing was performed by the Duke CGCB core on a rapid HiSeq 2500 instrument. All analysis was performed using the BD-generated analysis pipeline.
