COVID-19 High-Depth Blood PAXgene RNAseq

The goal of the project is to perform RNAseq from blood of COVID-19 patients or controls. Blood was collected into PAXgene tubes and sequenced at Ambry Genetics. Patients were consented and samples collected as approved by the Spectrum Health IRB. Patients that tested COVID-19 positive by PCR test had 2mL of blood drawn into PAXgene Blood RNA tubes (PreAnalytiX). RNA was isolated from PAXgene Blood RNA tubes using Thermo MagMAX Isolation Kit on Thermo King Fisher Presto. RNA yield was quantified using M200 Infinite (Tecan) and sequencing libraries prepared using a Globin-minus, RiboErase, RNA HyperPrep Kit (KAPA Biosystems). Whole transcriptome sequencing was done using 150 bp, paired-end reads on Illumina NovaSeq6000.
