The goal of this study is to analyze the effect of atp-citrate-lyase (ACLY) knock-down on gene transcription of bone marrow-derived monocytes (BMMs) with RANKL stimulation. BMMs with or without ACLY knock-down were stimulated with RANKL/MCSF for 3d and then total RNA was isolated and RNA-sequencing (RNA-seq) was performed, gene alteration with a fold change greater than 2 and adjusted p value less than 0.05 was identified as statistically significant. The experiment group contains two samples where murine BMMs isolated from two independent donors were infected with adenovirus encoding shRNA against ACLY and confirmed knock-down of ACLY; the control group contains two samples where murine BMMs isolated from two independent donors were infected with adenovirus encoding scrambled shRNA.
