In order to investiage the morphogenetic control of early tube development in the Drosophila melanogaster embryo we performed single-cell-RNAseq-analysis of FACS-isolated early salivary gland placodal cells (stage 10-12), making up less than 0.4% of cells within the embryo to look for early tubulogeneic signatures. In order to identify salivary gland specific signals from epidermal signatures we also performed single-cell-RNAseq-analysis on FACS-isolated epidermal cells in stage 10-12 Drosophila melanogaster embryos for the purpose of differential expression anaylsis. Two batches of salivary gland (SrcGFP) and one batch of epidermal tagged cells (ArmYFP) were obtained and isolated by flow cytometry from Drosophila embryos stage 10-12, and analysed by single cell sequencing.
