Based on the average values of the leaf size-related phenotypic data in three environments, 16 families with significant differences in leaf size were selected from the F2 population to construct two extreme mixed pools. Among them, the 8 families with larger leaves constituted the large-leaf pool (B), and the 8 families with smaller leaves constituted the small-leaf pool (L). The 16 selected F2 families were cultivated and grown according to the above method, and at 30 days, the terminal leaves at the same relative spatial position of each family were collected. To ensure equal total RNA in the two extreme mixed pools, 0.6 g of leaves were weighed from each family. The leaf samples from 8 large-leaf and 8 small-leaf F2 families were separately mixed to form the B pool and the L pool, and they were ground in liquid nitrogen using a mortar and pestle.
