Aseptic loosening caused by wear particle-induced osteolysis (WPO) is a major complication limiting the longevity of joint implants. This study aims to explore a "proactive preventive" design strategy that not only reduces wear particle generation but also mitigates their harmful immune effects, thereby preventing WPO. As a proof-of-concept, distal femoral tissues from mice implanted with bone screws and exposed to different particle types (none, polyethylene, or ceria nanozyme-engineered UHMWPE) were analyzed by transcriptome sequencing. This approach provides insights into both the mechanical and biological mechanisms underlying the prevention of implant failure.
