Bulk RNA-sequencing based transcriptome profiling of human AgRP and POMC neurons isolated by IHC/LCM-Seq

The goal of this project was to characterize the transcriptome profiles of human AgRP and POMC neurons, key regulators of energy homeostasis. To achieve this, we adapted our immunoLCM-Seq method for post-mortem human brain tissue. The technique labels targeted neurons using immunohistochemistry, and newly developed RNA preservatives to maintain RNA integrity throughout the entire process. Each transcriptome profile contains at least 15,000 individual transcripts expressed in either cell type. This approach also enables direct comparison between these two crucial neuronal populations, allowing the identification of differentially expressed genes.
