Comparison of transcriptome between vector-transduced (control) and TIFAB-transduced murine MLL-AF9 leukemia stem cells Leukemia stem cells (defined as lin−c-kit+Sca1−CD16+CD34+) were sorted by FACS from mice receiving MLL-AF9-transduced Kit+Sca1+Lin- bone marrow cells (isolated from NIKERT2 mice) and retrovirally transduced with empty vector (Con) or TIFAB (TIFAB). The transduced cells were sorted and injected into sublethally (6.5Gy) irradiated recipient mice, then leukemia stem cells from full-blown leukemic mice were isolated and used for RNA sequencing analysis.
