10 000 seeds of Arabidopsis reporter line (ProUBQ10:GCaMP3) were EMS-mutagenized (0.4% v/v) and M2 descendants were screened for their ability to sense E-2-hexanal. Mutants were selected and backcrossed to the parental line (ProUBQ10:GCaMP3) several times up to BC1F3 in order to confirm that a single mutation was at play. DNA of hexanal response mutants were identified and a pool of their DNA sent for sequencing. DNA from plants from the same generation with a hexanal response were also sent for sequencing to use as controls.
