Mast cells are implicated in the pathogenesis of rosacea; however, the therapeutic potential of tranilast, a mast cell membrane stabilizer, remains unexplored. This study aimed to evaluate the clinical outcomes and tolerability of tranilast monotherapy and its combination with minocycline in patients with rosacea. All enrolled patients were randomly assigned to receive tranilast, minocycline, or a combination of both. Tranilast (0.1 g, three times daily) and minocycline (50 mg, once daily) were administered for 12 weeks, with follow-up every two weeks.
Supplementary Figure 1 showed the participant flowchart. Supplementary Figure 2 showed the comparisons of the erythema index, melanin index, hydration, and transepidermal water loss (TEWL) before and after treatment in each group. Supplementary Figure 3 showed the comparisons of clinical symptom scores (redness, warmth, tingling, and itching) before and after treatment. Supplementary Figure 4 showed the comparisons of the GFSS score, lesion count, and RosaQoL scores, as well as post-treatment patient satisfaction across the three groups.
