Intracellular proteobacterial pathogens are some of the closest extant relatives to the ancestral protomitochondrion. Studying these diverse pathogens during infection of their hosts sheds light on characteristics of the common ancestor and gives deeper insight into their mechanisms of pathogenesis in modern diseases. Transcriptomic approaches provide global insights into intracellular acclimatization but are limited by excess host RNAs. We sequenced and assembled the genome of an aquatic Gammaproteobacterium and a common fish pathogen, Edwardsiella piscicida EtPo1, using long read technology, and we developed a method to enrich bacterial RNA from infected zebrafish, Danio rerio, BRF41 fibroblasts using magnetic nanoparticle technology.
