This study was designed to evaluate the transcriptomic alterations mediated by cisplatin exposure in a human cancer xenograft mouse model. The tumor samples from xenograft mice with or without cisplatin treatment were applied to transcriptomic analysis. 6-week-old athymic nude mice were subcutaneously injected with 1x10e6 human carcinoma cell line KB-3-1. The xenograft mice were randomly divided into 2 groups (10 mice/group) and were either treated with 0.5 mg/kg of cisplatin or PBS at day 5 and 5 mg/kg of cisplatin or PBS at day 10 after the xenograft. Tumors were collected at the experimental endpoint at day 20, and three tumors from each group were randomly selected and applied for RNA isolation and poly-(A) RNA sequencing.
