To investigate the antitumor mechanisms of the dual EP2/EP4 antagonist TP-18, Tumor-associated macrophages were isolated from CT26 tumors, with or without TP-18 treatment, and subjected to RNA-seq–based transcriptomic profiling. Female BALB/c mice (6–8 weeks old) bearing subcutaneous CT26 tumors were treated with either vehicle or TP-18 (50 mg/kg, QD, orally) for 3 weeks. Following treatment, mice were euthanized via cervical dislocation, and subcutaneous tumors were harvested and processed into single-cell suspensions. CD11b+F4/80+CD206+ TAMs were isolated using Fluorescence-Activated Cell Sorting. Total RNA was extracted using TRIzol reagent and stored in liquid nitrogen for subsequent sequencing.
