The termination of floral meristems is regulated by the MADS domain transcription factor AGAMOUS (AG) by preventing the maintenance of the H3K27me3 mark along the KNUCKLES (KNU) coding sequence. After 2 days of cell divisions, KNU transcription is activated prior to floral meristem termination. However, how many other downstream genes are temporally regulated by this intrinsic epigenetic timer and what kind of function those genes have are not known yet. Here, we identify AG direct targets regulated through cell cycle-coupled H3K27me3 dilution in a genome-wide manner. As seen in biotimer genes, such as KNU, AT HOOK MOTIF NUCLEAR LOCALIZED PROTEIN (AHL18) and PLATZ10, expression timing was correlated with the length of H3K27me3-marked region. Thus, we examined AHL18 binding in a genome-wide manner by ChIP-seq.
