We report that the addition of high amounts of amino acids, combined with the overexpression of transcription factors, results in a shift of the global transcriptome of both stem cell derived hepatocytes and HepG2 cells towards the profile of primary human hepatocytes, Overall design: HepG2 cells were either grown for 5 days in standard DMEM 10% FBS medium, or in medium suppelmented with high amounts of amino acids. Stem cell derived hepatocyets were differentiated towards the hepatic lineage without (HLC) or with the overexpression of 3 (HC3X) or 6 (HC6X) transcription factors, and this in standard medium or in medium supplemented with high levels of amino acids.
