The main hypothesis in the GOLD study is that specific patterns of the gut microbiome composition will be associated with diabetes, pre-diabetes and glycemic traits among Latino adults. The analysis and sequencing of the gut microbiome (GMB) will be performed at UCSD in the Knight Lab. Background information HCHS-SOL: 16,415 cohort members were recruited during 2008-11 and will complete six-year follow-up examinations during 2014-17. Measurements included fasting plasma glucose (FPG) and a 2h oral glucose tolerance test (OGTT), in addition to other laboratory (e.g., HbA1c, insulin), clinical (e.g., adiposity) and behavioral (e.g., diet, exercise) measures relevant to diabetes. Stool samples were collected from 2,000 subjects at all four HCHS-SOL sites (Miami, San Diego, Chicago and the Bronx area of New York). Analysis of the GMB with dietary data, and clinical data from HCHS-SOL will be performed on coded data at the Knight Lab using bioinformatics pipelines developed in his lab that will generate functional gene groups for each microbiome. Designed in the setting of a large, population based sample, HCHS-SOL is uniquely suited to discern ethnic differences in GMB makeup and to study the association of GMB with diabetes across birthplace/national background groups.
