To determine which GLT-regulated genes might be affected by compound D, we performed an RNA-sequencing analysis of rat islets treated ex-vivo with or without GLT and compound D for 3 hours and 24 hours. Compound E (related inactive compound) was used to account for non-specific scaffold effects. Overall design: Islets from 8 rats were pooled together and divided into 8 groups: 3hr GLT (27mM glucose, 300uM PA); 3hr control (11mM glucose, no PA); 24hr GLT;24hr control; 3hr GLT + cpd D 300nM; 24hr GLT + cpd D 300nM; 3hr GLT + cpd E 300nM; 24hr GLT + cpd E 300nM. After treatment islets were collected for RNA prep; Experiments were repeated 3 times (3 separate isolations)
