Spatial and single-nucleus transcriptomic profiling of CHB liver biopsies

A modified spatial transcrptomics workflow was used to profile liver tissues to compare viral and host-transcript profiles Overall design: The assay was tested on H&E stained FFPE samples from: 1) HBV infected uPA/SCID humanized mouse livers (one vehicle treated and one Tenofovir alafenamide treated), 2) cell pellets from PLC/PRF/5 cell line and primary human hepatocytes which where unifected or infected with genotypes C and B versions of HBV, and 4) commercially procured liver biopsy samples (2 healthy donors, 4 chronic HBV patients)
