The present study aims to synthesize zinc oxide nanoparticles using propolis extract and evaluate its effect on zebrafish immune response following caudal fin amputation model on zebrafish (Danio rerio). The phytochemical composition of extract was identified using Gas Chromatography Mass Spectrometry (GC-MS). The synthesized Pro-ZnO NPs were characterized using UV-Vis spectrophotometer, Fourier transform infrared (FTIR) and transmission electron microscopy (TEM) analysis. The toxicity of Pro-ZnO NPs was evaluated in zebrafish larvae, with the LC50 values of 4 mg/L which classified as toxic. Male zebrafish (n = 5 per group, triplicate) underwent caudal fin amputation and were divided into two groups to assess their immunological response: a control group and a Pro-ZnO 2 mg/L treatment group for a duration of four days. Total RNA was extracted from the caudal fin blastema four days post-amputation (dpa), and subsequently subjected to strand-specific mRNA sequencing using Illumina Platform. The principal component analysis (PCA) yielded a clear delineation between the ZnO-treated group and the control group, thereby indicating a distinct grouping pattern.
