A novel colon cancer-specific membrane protein, TMEM180, was found, and it was found to be expressed as 12-transmembrane topology and upregulated at low oxygen. We have already established a humanized monoclonal antibody and are aiming for clinical trials. Before that, it is necessary to clarify the function of this molecule in cancer. Cell proliferation ability, oxygen consumption, phosphorylation proteomics, metabolomics, and next-generation sequence were performed between human colon cancer cells SW480 and its TMEM180 knockdown cell. It was revealed that TMEM180 promotes the growth of colon cancer. The molecule was independent of oxygen consumption, and was almost unrelated to phosphorylated protein expression. In the glycolysis system, a big difference was recognized between the two. It was found that it promotes nitric oxide synthesis, which suggests that it promotes glucose metabolism and glutamine metabolism and is involved in cancer growth. This study supports the clinical development of anti-TMEM180 antibody.
