Analysis of three subsets of monocyte-derived macrophage generated using various differentiation cocktails: M1 (IFN-γ+LPS), M2a (IL-4+IL-13) and M2c (IL-10). Overall design: Untouched monocytes were isolated from the PBMC of two healthy donors. Monocytes were then differentiated to M0 macrophages upon culturing monocytes with M-CSF (100ng/ml) for 6 days. On day 6 M0 macrophages were further stimulated with differentiation cocktail for 24 hour to obtain three different subsets of monocyte-derived macrophages namely M1 (stimulated with IFN-γ+LPS), M2a (stimulated with IL-4+IL-13) and M2c (stimulated with IL-10).
