The goal of this study is to determine the onset and longitudinal contribution of hematopoietic stem cells to the blood after transplantation, engraftment, and reconstitution. To address this question, lentivirus gene-modified hematopoietic stem and progenitor cells were transplanted into myeloablated nonhuman primates and clones tracked over time using retroviral integration site analysis. The other goal is to identify transcriptionally distinct subsets within human GCSF-mobilized hematopoietic stem and progenitor cells (CD34+), determine the portion of primitive multipotent hematopoietic stem cells, and identify signatures associated with previously described functionally-distinct phenotypes.
