Regulating the community structure of gut microbiota is vital to human health. We proposed a new strategy to swiftly adjust the microbiota by the delivery of supplement of glucose to colon based on pH-sensitive embedment and spray drying. It may be called as delivering ‘glucose parcel’ to the right place. The effects of this act on gut microbiota have been detected by both in vitro and in vivo experiments in rats. In vitro fecal fermentation showed 50 mg/mL glucose enhanced proliferation of Lactobacillus and Bifidobacterium, having suppressed Romboutsia. In vivo experiments revealed feeding 0.1 g/d (body weight: 180 ± 20 g) for 4 days of the glucose parcel for rats increased relative abundance of Lactobacillus to 37.24% in transverse colon, and enriched the beneficial genus Lachnospiraceae_NK4A136_group and norank_f_Muribaculaceae. Untargeted metabolomics analysis indicated that supply of the exogenous glucose mainly activated metabolic pathways related to antibiotics and neurotransmitters. These responses seem not built on a significant production of short chain fatty acids. No significant adverse effect on blood glucose or liver function was observed after a 10-day feeding of the glucose parcel to rats. These data suggest that the targeted delivery of ‘glucose parcel’ to colon may be a promising and cost-effective strategy to modulate gut microbiota. Indeed, this may inspire other types of ‘parcels’ to be created for specific purposes of modification of microbiota.
