Amplicon sequencing of fungal and bacterial communities in surface soil horizons in the Ogasawara islands, Japan

The goal of this project was to understand microbial island biogeography and endemism of soil microbes. The study was conducted on two different islands in the Ogasawara Islands, Chichijima (n=2) and Hahajima (n=1), located in the northwestern Pacific Ocean. We used amplicon-based sequencing of bacterial 16S rRNA gene V4 region (515F/806R) and fungal ITS2 region (gITS7/ITS4ngs) to characterize the microbial community in O- and mineral soil horizons. Sequencing for bacterial and fungal communities was performed on a MiSeq platform (Illumina, San Diego, CA, USA) using 2 Ã? 250 bp and 2 Ã? 300 bp paired-end reads, respectively.
