Our study revealed that POMC neuron-specific knockout of TOX3 significantly exacerbates high-fat diet-induced obesity and related metabolic disorders. Further investigation demonstrated that TOX3 knockout markedly reduces POMC neuronal activity and POMC expression. To elucidate the molecular mechanism by which TOX3 regulates POMC neuronal function, we performed single-nucleus RNA sequencing on the hypothalami of high-fat diet-fed control mice (LoxP) and POMC neuron-specific knockout mice (PTKO).
