Chromatin accessibility is a key factor involved in gene transcriptional expression and is critical for tumor occurrence and development. Demethyltransferase regulates chromatin conformation by changing histone modification, resulting in the activation or inhibition of multiple gene transcription affecting tumor progression. As a small molecular inhibitor of demethyltransferase, IOX1 has certain chemotherapeutic activity for many kinds of tumors. However, it is unknown whether IOX1 has a potential role in regulating radiosensitivity and how it regulates global transcription in lung cancer. In this study, we found that chromatin accessibility was significantly altered after the treatment of IOX1 by the detection of ATAC-seq. There was a positive correlation between the changes in promoter accessibility and transcriptional expression of DNA damage-repair-related genes.
