Expression of GPR153 is upregulated in response to damage both in smooth muscle cells and endothelial cells.We found knockdown of GPR153 in SMC reduced the ligation-induced neointima formation, and GPR153 defeciency in EC protect the mice from neuroinflammation. The mechanism of orphan GPCR GPR153 in regulating cell signaling were studied in SMC, EC and HEK293t cell lines. HEK293t cells were transfected with scramble siRNA or GPR153 siRNA, and then transfected with empty vector or GPR153-Myc plasmid. Control group (scramble siRNA, empty vector), GPR153 knockdown group (GPR153 siRNA, empty vector) and GPR153 overexpression group (scramble siRNA, GPR153-Myc plasmid) were sent for mRNA sequencing.
