Transcriptional profiling of sorted THP1 cells infected with wild-type Mtb (H37Rv) or ESX-1-mutant Mtb

THP1 cells were infected with wild-type or ESX-1-mutant Mtb expressing GFP. Cells were then sorted into infected and bystander populations, and RNAseq was performed. PMA-differentiated THP1 cells were infected with wild-type H37Rv-GFP or ∆ESX-1-Rv-GFP at an MOI of 10:1. 4 hours were allowed for phagocytosis, then cells were washed with PBS and the media was replaced. Cells were flow sorted into infected (GPF+) and bystander (GFP-) populations at 2, 4, 8, and 16 hours after the wash. Total RNA was isolated and bulk RNA sequencing was performed.
