This work investigated multivalent HIV-1 Env immunization strategies and provides a novel methodology for screening guinea pig B cell receptor antigen specificity at a high throughput level using LIBRA-seq. B cells specific to DNA-barcoded envelope glycoproteins of HIV-1 strains were sorted from the splenocytes of two separate guinea pigs. Both guinea pigs were administered with a mix of three strains of HIV-1 Env immunogen (286.36, 5768.04, and KNH1209.18) at three time points, each four weeks apart. Cells were distributed into microfluidic droplets containing unique cell-specific oligonucleotides using the 10X Genomics Chromium Controller. cDNA libraires from single cells were further amplified with a custom mix of BCR enrichment primers specific to guinea pig IgG genes.
