Cellular collectivity and its effect on T cell memory formation. Two experiments were performed: 1) 20160314. Gene expression of early differentiated CD62Lpos and CD62Lneg CD4 T cells. 2) 170213. Gene expression of CD4 T cells cultured in high and low initial concentrations over time. And, early CD4 T cell memory differentiation Overall design: Two experiments were performed: 1) 20160314. naïve CD4 T cells were cultured at an initial concentration of 2.5E5 cells/ml. Cells were activated using antiCD3/antiCD28 coated beads. Cells were cultured in 200ul complete RPMI in 96-well plates. At the indicated times cells were sorted to CD62Lpos and CD62Lneg populations. 2) 20170213. naive CD4 T cells were cultured at either 6.25E4 or 1E6 cells/ml. Cells were activated using antiCD3/antiCD28 coated beads. Cells were cultured in 1ml complete RPMI in 24-well plates. at the indicated times CD4pos cells were sorted. additionally, naive CD4 cells were cultured at an initial concertation of 2.5E5 cells/ml similarly to the first experiment, and sorted at the indicated times to CD4posCD62LposCD44pos or CD4posCD62LnegCD44pos populations.
