In this study we show that human phagocytes carrying the nonsense mutation Y430* in Perforin-2 (MPEG1) have significantly reduced capacity to kill intracellular Salmonella enterica serovar Typhimurium and Yersinia pseudotuberculosis.
FIG1CD. S. Typhimurium or Y. pseudotuberculosis were used to infect Perforin-2 (Y430*) haploinsufficient neutrophils and macrophages or wild-type controls. A/G indicates age and gender matched control. Gentamicin was used to eliminate extracellular bacteria. Intracellular bacteria were enumerated at the indicated times post infection. The data sheet contains the raw data (CFU/ml) and after its transformation to Log2 Fold Change. Log2FC = log2(CFU at time X) – log2(CFU at initial time).
FIG2B. Interferon-gamma induces the expression of MPEG1 in derived haploinsufficient (Y430*) macrophages. Data was normalized to GAPDH expression and is presented relative to untreated macrophages.
