Dataset for the project "Targeting default mode network dysfunction in persons at risk of Alzheimer's disease with noninvasive techniques" (GR-2018-12368250)

Specific Aim 1: To test the feasibility of DMN modulation in subjects at risk for AD. Elderly subjects carrying the APOE4 allele will be included and randomized to two stimulation protocols (real versus sham rTMS). The effect of DMN stimulation will be assessed with rs-fMRI and TMS-EEG.

Specific Aim 2: To assess the cognitive effect of DMN modulation in subjects at risk for AD. The DMN is closely linked to memory and autobiographic functions. To evaluate whether DMN stimulation is a memory enhancer, subjects will perform an associative memory fMRI task before and after stimulation.

Specific Aim 3: To elucidate the mechanisms underlying DMN modulation. Proposed mechanisms of action of neuromodulation include neuronal activity and axonal activation. We will investigate these mechanisms in vivo at high spatial and temporal resolution using task fMRI and TMS-EEG, respectively, for neuronal activity; DTI and TMS evoked potentials (TEPs) for axonal activation. Since baseline connectivity might drive response to stimulation, we will also test whether baseline measures can predict response to rTMS.
