To clarify mouse PPARA autoinduction and hepatocyte proliferation signaling pathways, genome-wide distribution of PPARA in the liver of PPARA agonist-treated mice compared to Vehicle was analyzed. Overall design: ChIP was carried out to pull down PPARA bound chromatin from nuclei isolated from Vehicle (Control)- or PPARA agonist (Wy14,643)-treated C57BL/6 mouse livers. This chromatin fraction was then digested using micrococcal nuclease to release mononucleosomes. DNA fragments purified from nucleosomes were then used to generate libraries and sequenced.
