Double strand break-induced gene conversion frequency.

Analysis of HR frequency induced by I-Sce1-induced DSBs in an artificial substrate. SCneo-puro was stably integrated into the OVALBUMIN locus of indicated genotypes by gene-targeting. 1.0×107 cells of each genotype were transfected with 30 µg of either I-Sce1 expression vector or pBluescript KS, and subsequently selected using a neomycin analog (G418). The ratio of recombination frequency was calculated by comparing the number of G418 resistant clones in the mutant cell lines to that in the wild-type cell line. Only results obtained using the I-Sce1 expression vector are shown.
