RNA-seq analysis was performed in BPDCN patient-derived cell line CAL-1 (WT and METTL3 KO) to analyze gene expression changes after loss of METTL3 concurrent with GSK595 treatment METTL3 KO was induced in CAL-1 cells with Tetracycline and cells were subsequently treated with DMSO (control) or GSK595 (0.31 uM) for 72 hours. Total RNA was extracted using miRNeasy Mini Kit. Strand-specific library construction and 150-bp paired-end sequencing with a sequencing depth of 130M reads on the NovaSeq6000 platform were performed at Macrogen.
