In order for bacterial pathogens such as the Group A Streptococcus (GAS) to obtain critical nutrients during infection, they must be able to sense and coordinately regulate genes in the host required for nutrient utilization. Two such regulatory pathways are RofA, which senses carbohydrates, and PdxR, which monitors vitamin B6 levels, in order to alter their uptake and metabolism in GAS. Using RNA-seq, we have compared the transcriptomes of GAS mutants in PdxR (Delta-pdxR) and RofA (Delta-rofA) to the wild type parental M1T1 invasive isolate 5448 under relevant conditions in vitro to better understand their regulons and their impacts on GAS pathophysiology.
