Class IIa histone deacetylase 7 (HDAC7) plays a significant role in regulating gene transcription in chromatin, despite its low deacetylase activity. HDAC7 overexpression is linked to poor clinical outcomes of colorectal cancer (CRC). We found that HDAC7 overexpression promotes CRC progression by downregulating ATF3 and activating oncogenic signaling pathways linked to cellular proliferation and survival, resulting in the modulation of apoptosis and cell cycle arrest genes such as Bcl-2 and p21, thereby inhibiting cancer cell proliferation. Taken together, our data provide a molecular framework for transcription factor regulated by HDAC7 in SW480 cell lines.
