Supporting Data and Analysis Pipeline for the OA Glutamine Metabolism Study

This repository contains the datasets, analysis scripts, and supplementary materials associated with the study titled “Identification and validation of a glutamine metabolism-related gene signature as a diagnostic and therapeutic target in osteoarthritis through integrated multi-omics analysis”. The study presents a multi-tissue, multi-omics investigation integrating GEO transcriptomic datasets from cartilage, synovium, subchondral bone, and peripheral blood to explore the role of glutamine metabolism in osteoarthritis (OA). Using a machine learning pipeline combining WGCNA and LASSO regression, we identified a core three-gene glutamine metabolism–related signature (F13A1, IRS2, RELA). The diagnostic performance of this signature was validated in an independent cohort (31 OA patients and 31 controls) using RT-qPCR and plasma metabolomics. This repository provides the processed data, scripts used for computational analyses, and supplementary files required to reproduce the results presented in the manuscript.
