In this study, we assess potential differences in mechanism of action for two PAHs, benzo[a]pyrene (BAP) and dibenzo-[def,p]chrysene (DBC), in a primary human 3D bronchial epithelial culture (HBEC) model based on short-term biosignatures identified from global transcriptional profiling. Overall design: Fully differentiated bronchial epithelial cells (n=4 replicates) were treated with 500 µg/ml (19.80 nanomoles) benzo[a]pyrene (BAP), 10 µg/ml (0.35 nanomoles) dibenzo[def,p]chrysene (DBC) or acetone vehicle control for 48 hrs and isolated mRNA were sequenced on Illumina HiSeq 3000.
