Transcriptomic Profiling on Kidneys of Mice after Cecal Ligation and Puncture (CLP) Compared to Sham Operation

Sepsis is a severe clinical syndrome related to an exaggerated host immune response to infection as well as systematic inflammation and serious tissue damage. Sepsis-associated acute kidney injury (SA-AKI) is one of the most frequent and serious complications of sepsis. This study aimed to identify new mechanisms in SA-AKI through transcriptome profiling (RNA-seq). A total of 5 C57BL/6 mice were randomly divided into 2 groups: CLP group (n=3), Sham group (n=2). In CLP group, we first anesthetized mice 2%~4% isoflurane. Under aseptic conditions, a 2-cm midline laparotomy was created below the diaphragm to expose the cecum. Two-thirds of the cecum were ligated with a 5-0 silk suture and punctured twice with a 22-gauge needle. The cecum was then squeezed gently to extrude a small amount of feces through the perforation site. Animals were resuscitated with 1 mL of saline subcutaneously after CLP. The procedures of the Sham group (controls) were the same as that of the CLP group, except for ligation and perforation. At 6 hours after CLP, the mice were sacrificed by neck fracture, and the kidneys were taken for RNA sequencing.
