This study uses ATAC-seq to profile chromatin accessibility in accessory gland tissue from multiple Drosophila species and accessions to investigate the regulatory basis of de novo gene origin. We generated ATAC-seq libraries from 2-day-old male accessory glands for five Drosophila melanogaster accessions (R304, R357, R360, R399, and R517), with three biological replicates per accession, three biological replicates of a single Drosophila simulans accession, and one biological replicate of a single Drosophila yakuba accession. The purpose of this study was to characterize natural variation in chromatin accessibility and to identify regulatory features associated with accessory-gland-expressed de novo genes.
