Quiescent p57+ cells located around +4 position of crypts serve as facultative intestinal stem cells that dedifferentiate in response to injury and contribute to post-injury regeneration. To investigate the population-wide transcriptomic changes in this process, we performed single-cell RNA-seq analysis on p57-Venus+ and p57-Venus- cells isolated from normal or injured intestinal crypts by using CEL-seq2 platform. This analysis uncovered that, during injury-induced reversion, p57+ cells go through a spatiotemporal reprogramming process that is characterized by both fetal-like reversion and metaplasia-like transformation of the transcriptomic signatures.
