This study adopted a single sample time series design, with 0h (pig1) as the baseline control and 12h (pig2), 24h (pig3), and 36h (pig4) as dynamic modeling nodes. Through multi-dimensional synchronous tracking of "cytokines+pathology+microbiota", the changes in intestinal microbiota under acute cytokine storm were accurately captured. Four Bama pigs were randomly divided into groups and underwent jugular vein catheterization to receive different intervention measures (1) The control group (n=1) underwent anesthesia catheterization treatment, and blood was drawn for testing at 0 hours; Subsequently, under deep anesthesia, circulatory failure was caused by intravenous injection of air, and the final organ (small intestine) was euthanized for sampling. A portion of the small intestine tissue was stored in liquid nitrogen and placed in a -80 ℃ freezer for analysis of gut microbiota. The remaining pathological specimens were fixed in formalin solution and subjected to pathological examination. (2) The experimental group (n=3) also underwent anesthesia catheterization treatment, and blood was drawn at 0 hours; Immediately proceed with D-GaLN modeling (extract the configured D-GaLN solution at a dose of 1.5g/kg and slowly push the mold through a dual chamber Arrow tube). Afterwards, at 12/24/36 hours, under deep anesthesia, circulatory failure was caused by intravenous injection of air, and the animals were euthanized. At the end of the day, organs were collected for pathological and intestinal microbiota testing, and tissues were collected, stored, and transported according to the same procedures and requirements as the control group; Blood cytokine testing was conducted every 12 hours during the survival period of Bama pigs. This study set up a sham operation control of jugular vein catheterization and infusion procedures, which helps to partially control the interference of the operation itself on the inflammatory response. All Bama pigs are sampled every 12 hours and euthanized at the endpoint to ensure that cytokine levels, intestinal pathological status, and microbiota composition at each time point have temporal correspondence, providing a basis for subsequent correlation analysis. The obtained data includes cytokines (IL-6/IL-10), intestinal pathological sections, and gut microbiota related data.
