The CRISPR-Cas system is a new DNA-editing technology and promise potential in many applications such as genome engineering and genetic screening. We generated a genome-wide lentiviral CRISPR guide RNA library for mouse. Using the library we generated mouse mutant ES cell libraries. The libraries in theory contain mutants for every gene and can be used for genetic screening. In this study, we want to find genes whose mutations confer resistance or hyper sentisitity to DNA damaging agents such as etoposide and cisplatin.
