To understand the transcriptional changes on cellular level within the murine intestinal epithelial tissue, we generated small intestinal organoids from wild type mice, co-cultured them with Treg cells +/- rIL10/rIFNg or anti-IFNg/IL10R, dissagregated the samples, sorted into single cells and profiled them by single-cell RNA-seq Overall design: To understand small intestinal epithelial normal tissue differentiation and growth during the presence of Treg cell and/or IL10/IFNg signaling, small intestinal organoids were generated and co-cultured with indicated wild-type Treg cells +/- anti-IFNg/IL10R or +/- rIFNg/rIL10. Medium was changed after 4 days, stimuli were removed and the organoids were dissagregated, sorted into single cells and profiled by single-cell RNA-seq
